中文摘要: 磁共振成像是一项全球每年应用超过6000万次的临床检查技术;钆配合物是一种磁共振成像中常用的造影剂。然而近期研究发现,使用钆配合物作为造影剂有可能诱发一种严重的皮肤疾病(肾源性系统纤维化)。进一步研究显示,钆配合物、铁蛋白、血清铁三者之间存在着可能的关联。为更好的理解这种关联性,从基础研究的角度出发探究钆配合物、脱铁铁蛋白、Fe2+三者之间的关联是很有必要的。由此本项目提出钆配合物复合对脱铁铁蛋白催化性能的影响及其机理研究。本项目将制备并表征一系列脱铁铁蛋白-钆配合物复合物,研究该复合作用对于钆配合物弛豫率和Fe2+矿化动力学的影响。本项目的特色之处在于巧妙设计了Tb3+荧光检测实验和双核铁过渡态瞬态吸收动力学实验这两个实验以确定钆配合物在脱铁铁蛋白中的结合位置,从而揭示钆配合物复合对脱铁铁蛋白催化性能影响的机理。本项目注重利用光学技术(包括吸收光谱、荧光光谱和瞬态光谱)解决一些特定问题。

英文摘要: Magnetic resonance imaging is a clinical examination technique that is annually utilized more than 60 million times worldwide. Gd chelates are a kind of popularly used contrast agents in magnetic resonance imaging. However, recent research discovered that the administration of Gd chelates as the contrast agent could cause a serious skin disease (nephrogenic systemic fibrosis). Further study revealed a possible association among Gd chelates, ferritin, and serum iron. To better delineate this association, it is necessary from the basic research point of view to investigate the association among Gd chealtes, apoferritin, and Fe2+. Therefore, this project aims to work on the effects of Gd chelate complexation on the catalytic properties of apoferritin and the mechanistic studies. Herein, a series of apoferritin-Gd chelate complexes will be prepared and characterized. The effects of the complexation on the relaxivity of Gd chelates and the kinetics of Fe2+ mineralization will be studied. A special and innovative contribution in this project is the smart design of two experiments -- Tb3+ fluorescence measurement study and transient absorption kinetics study of the di-iron transition states.
