中文摘要: 肽聚糖识别蛋白(PGRPs)是一类能特异性识别肽聚糖和含肽聚糖细菌的模式识别受体蛋白,在固有免疫应答中发挥重要作用。本项目以长牡蛎为研究对象,拟采用生物信息学、分子生物学和免疫学技术,克隆长短型 PGRPs 基因,确定其系统进化关系;在个体水平研究这些基因在病原相关分子模式刺激下的表达特征;体外重组各类 PGRPs,比较它们的免疫学活性差异,探讨其在整个免疫应答中的分工与协作关系;定点突变特定氨基酸,阐明它们与某种免疫学活性间的关系;检测相关信号通路中的基因表达及免疫学指标的变化,确定长牡蛎中由PGRPs介导的免疫应答途径;沉默 PGRPs 在体内的表达,明确其在长牡蛎抵御病原微生物侵袭中的作用。该项目将为进一步揭示 PGRPs 的进化脉络提供证据,为系统深入了解软体动物各类 PGRPs 的功能、分工和作用机制提供有力依据,并为丰富和发展海洋无脊椎动物免疫学内容提供新的思路和理论支持。

英文摘要: Peptidoglycan recognition proteins (PGRPs) are a class of pattern recognition receptor proteins,which can recognize peptidoglycan and bacteria containing peptidoglycan specifically, and play an important role in the recognition and regulation of the innate immune response. In the present project, Crassostrea gigas is selected as the research object, and the techniques in bioinformatics, molecular biology and immunology will be used to (1) clone the full-length cDNA of S-type and L-type PGRPs, and analyze their phylogenetic relationship; (2) investigate the expression pattern of these PGRPs after oyster are stimulated by pathogen-associated molecular patterns; (3) recombine these PGRPs and compare their immune functions to investigate the division of labor and collaboration in the immune response; (4) mutate the key amino acids of the PGRPs and clarify their relationship with immunological activity; (5) investigate the change pattern of related gene expression and immunological indexes in the signaling pathway and determine the possible immune activation pathway mediated by PGRPs; (6) silence the expression of these genes and reveal their roles in immune defense against pathogens.
