中文摘要: 上皮间质转化是上皮细胞来源的恶性肿瘤细胞获得迁移和侵袭能力的重要生物学过程,因其在肿瘤转移中的作用成为研究热点。miR-200家族是调控肿瘤上皮间质转化的最主要miRNAs 之一,TGF-β及ZEB均为其作用靶点。片仔癀是传统名贵中成药,临床实践表明其对结肠癌具有显著疗效。课题组前期研究表明片仔癀有显著下调结肠癌细胞miR-200a的作用,本课题组在前期研究基础上以人结肠癌肝转移动物模型,以及体外培养人结肠癌细胞HT-29、CT26等为研究平台,采用激光共聚焦、transwell、异质粘附、实时荧光定量PCR、western-blotting等实验方法,系统地研究片仔癀对结肠癌上皮间质转化的影响,阐明片仔癀对结肠癌的治疗作用及其分子机制和作用靶点,以期为临床片仔癀治疗结肠癌提供进一步的理论根据。

英文摘要: Epithelial-mesenchymal transition(EMT) is an important biological process, where malignant tumor cells from endothelial cells gain more ability of migration and invation. People have been paid more attention to EMT for its role in tumor metastasis. miR-200 family is one of miRNAs which regulate tumor epithelial-mesenchymal transition, both TGF-β and ZEB are its targets. Pien Tze Huang , a well-known traditional Chinese formula , has been demonstrated to be clinically effective in the treatment of colorectal cancer (CRC).We have demonstrated that Pien Tze Huang can down-regulted miR-200a significantly in CRC cells. Based on this results,we will construct CRC metastasis animal model and culture HT-29 and CT26 cells. Confocal laser scanning microscope, transwell, Heterogeneous adhesion experiment, real-time PCR and western-blotting were used to research the effect of Pien Tze Huang on epithelial-mesenchymal transition in CRC, and liiuminate the mechanism and effective target by which Pien Tze Huan treat CRC.
