中文摘要: 肺癌是世界上发病率和死亡率最高的恶性肿瘤之一,大部分患者死于肺癌的复发和转移。血液中循环肿瘤细胞(Circulating Tumor Cells,CTCs)直接来源于肿瘤组织,CTCs的检测可用于肿瘤检测、疗效评价、复发及转移的监测。CTCs在血液中含量极少,建立高灵敏、高特异的CTCs检测及鉴定体系,是一大研究热点。但大部分研究报道是集中在改进CTCs检测技术方面的,对于检测到的CTCs分子表型鉴定的报道极少。对CTCs进行分子表型鉴定要比仅仅对CTCs计数更有临床意义。本项目通过研究磁珠捕获法及微流体芯片微过滤管道两种不同CTCs富集分离方法得到的CTCs表面的分子及表面标记特性,主要包括间质、上皮、肿瘤干细胞等标志物及基因突变、基因图谱情况,找出与肺癌疗效、复发转移及个体化治疗密切相关的CTCs特性,构建高灵敏、高特异的CTCs鉴定新方法,建立有效的个体化治疗指导及肺癌复发监测体系.

英文摘要: Lung cancer is one of the most malignant tumors with high morbidity and mortality in the world. Most of cancer patients died of recurrence and metastasis of lung cancer. Circulating tumor cells (CTCs) in the peripheral blood detach from the primary tumour directly. Detection and analysis of CTCs can provide information on patient prognosis, progression and treatment efficacy. CTCs are present in the bloodstream at a low concentration. Hence, methods for detecting CTCs need to be sensitive, reliable and specific. However, most of the technologies have improved the recovery of CTCs from patients, the characterization of these cells is scarce. Molecular and phenotypical characterization of the CTCs is more important than just counting of these cells. This project is about the molecular and phenotypical characterization of the CTCs enriched with immuno-magnetic beads or microfluidic chip. Mesenchymal, epithelial, cancer stem cell biomarker and gene mutation or gene map will be annalyzed for these CTCs. The subtype of CTCs correlating with patient prognosis, progression and treatment efficacy will be detected and identified with this study.
