中文摘要: 中华绒螯蟹是我国重要的经济甲壳动物之一,在正常情况下,一定数量功能性精子的形成对该物质的繁殖至关重要。生精细胞凋亡是控制精子质量的重要途径之一,但目前动物精子质量下降是普遍问题,原因未知。尽管p53被证明通过参与自发性和诱导性细胞凋亡调控精子发生过程,但迄今大多数对该调控机制研究仅限于哺乳动物。p53是否及如何参与和调控中华绒螯蟹生精细胞的凋亡至今无人问津。本课题组已经从中华绒螯蟹精巢中克隆出p53基因,并对其mRNA表达和分布做了初步研究。在现有结果基础上,我们认为p53可能会通过内源性或者外源性细胞凋亡路径参与中华绒螯蟹生精细胞的凋亡过程,p53蛋白的泛素化调控可能和p53调节正常的精子发生及生精细胞凋亡密切相关。本项目拟用基因克隆、原位杂交、定量PCR、免疫共沉淀、免疫荧光、免疫电镜、TUNEL、Annexin V等研究技术验证上述假设,其结果将揭示中华绒螯蟹生精细胞凋亡的可能机制。

英文摘要: Chinese mitten crab, Eriocheir sinensis, is one of the important economic crustacean in China. To keep its high efficient reproduction, high quality and large number of functional sperm are quite essential.At present, no investigation has been done to elucidate the mechanism for controlling sperm quality and number during spermatogenesis of Chinese mitten crab. Though p53 was proved to be involved in the regulation of spermatogenesis by participating in spontaneous germ cell apoptosis and stress-induced germ cell apoptosis, those studies on p53 were only focusing on spermatogenesis in mammals. Whether and how p53 regulate germ cell apoptosis during spermatogensis of Chinese mitten crab hasn't been investigated, and elucidation of this problem showed a great theatrical importance and application value to future detailed clarification of spermatogenesis in this species. Our group had cloned p53 gene in the testis of Chinese mitten crab, and we also performed in vitro hybridization to detect p53 mRNA expression pattern during spermiogenesis in this species. Based on our prelimilary study, we thought that p53 may take part in the spontaneous germ cell apoptosis and stress-induced germ cell apoptosis probably occurred during the spermatogenesis of Chinese mitten crab.
