中文摘要: 原因不明性复发流产(URSA)是妊娠常见并发症,严重影响育龄妇女身心健康。但病因不明确,治疗效果不满意。绒毛外滋养细胞(EVTs)表达的HLA-E,可影响母胎界面通过CD94/NKG2受体进行的重要信号传递,其在调节母-胎间细胞相互作用及形成有利于维持妊娠的母胎界面免疫环境中发挥的作用受到关注。我们前期研究基础和国内外文献报道提示:孕激素可上调EVT体外模型JEG-3和早孕滋养细胞的HLA-E表达; HLA-E可能参与介导孕激素对母胎界面的免疫调控,从而有利于妊娠的维持。本项目拟结合miRNA芯片、生物信息学技术,采用自然流产动物模型,应用基因转染、慢病毒载体和 RNA干扰技术和荧光酶报告基因等方法,探讨HLA-E介导孕激素对母胎界面的影响及miRNA调控作用,为进一步阐明URSA的病因提供新的线索,并为开拓治疗URSA新途径提供理论依据,以期更好地应用于临床。

英文摘要: Unexplained recurrent abortion (URSA) is a common pregnancy complication women in fertile age suffering. The mechanism of URSA is unclear and the effects of treatments now available are unsatisfactory in the whole. As a major ligant of CD94/NKG2 receptors on decidual NK cells, HLA-E on extravillous trophoblasts (EVTs) may have effect on pregnancy maintenance by regulating fetal-maternal interface through interaction with CD94/NKG2 receptors. And the role of HLA-E in EVTs is causing big concerns. Our previous research work and studies by other researchers indicated that progesterone up-regulates HLA-E expression in EVT model and trophoblast cells in first trimester, and HLA-E may play an role in immunoregulation by progesterone at fetal-maternal interface. In this study, we will study the impact of HLA-E on immunoregulatory function of progesterone at fetal-maternal interface and its miRNA control using miRNA microarray, Bioinformation Technology, mouse miscarriage model, transfection, lentiviral vector and RNA interference technology, aiming to elucidate the mechanism of URSA and provide clues to effective new treatment to URSA.
