中文摘要: GM3是一种含有唾液酸的三糖抗原,在许多肿瘤细胞中广泛表达,因此成为设计治疗性肿瘤疫苗的热门靶标。然而,在基于GM3的肿瘤疫苗的开发上存在的一个严重问题是GM3的抗原性极差。前期的研究显示糖复合物GM3NPhAc-KLH具有很好的免疫活性,另外四种苯环上取代的糖复合物的免疫活性比GM3NPhAc-KLH更强,而GM3NPhAc-MPLA的免疫活性亦比GM3NPhAc-KLH 的免疫活性强,本项目拟在这些研究基础上,通过进一步的结构优化改造,合成四种肿瘤疫苗GM3NMePhAc-MPLA、GM3NMeOPhAc-MPLA、GM3NAcPhAc-MPLA、GM3NClPhAc-MPLA,寻找高效、低毒、并有较好药代参数的有继续开发价值的新药候选前体分子,为后续研发具有自主知识产权的修饰型GM3-MPLA糖复合物肿瘤疫苗奠定良好的实验基础。

英文摘要: GM3, a sialylated trisaccharide antigen expressed by a number of tumors, is an attractive target in the design of therapeutic cancer vaccines. However, a serious problem associated with GM3 is that it is poorly immunogenic. Previous studies had shown that glycoconjugates GM3NPhAc-KLH had the advanced immune activity, on the other hand , the immune activity of the other four benzene ring substituted glycoconjugates greater than GM3NPhAc-KLH and GM3NPhAc-MPLA immune activity was more greater than GM3NPhAc-KLH. This project is to be optimized through further structural transformation on the basis of these studies, synthesize four kinds of tumors vaccine GM3NMePhAc-MPLA, GM3NMeOPhAc-MPLA, GM3NAcPhAc-MPLA, GM3NClPhAc-MPLA, looking for efficiency, low toxicity, andbetter pharmacokinetic parameters of new drug candidates continue to develop the value of the precursor molecules, a modified type of independent intellectual property rights of the GM3-MPLA sugar, complex tumor vaccine to lay a good experimental basis for subsequent research and development.
