中文摘要: 生物信息学分析和RT-PCR验证,水稻基因组中存在8个vdac基因,分别定位于第1、3、5、9号染色体上。本项目对水稻基因组中vdac基因家族的8个候选基因转录表达的时空特异性进行分析,研究不同vdac基因与水稻生长、发育间的关系;通过克隆全长cDNA的序列分析和蛋白产物的预测,比较不同VDAC蛋白特异功能域的异同;以不同VDAC特异结构域制备的抗体,对从水稻不同组织中通过分级分离得到的各种细胞器及其蛋白质提取物进行特异性免疫杂交分析,研究不同vdac基因表达的特异性和亚细胞定位;对vdac特异性表达组织的线粒体膜渗透性及渗透性转换特性的分析,研究不同vdac基因表达与线粒体功能间的关系;采用RNAi技术敲除1-2个特异性表达的vdac基因,探讨vdac基因家族不同成员在水稻生长发育中的作用。本研究对于阐明水稻vdac多基因家族存在的必要性及其生物学功能,指导其实际应用有重要意义。

英文摘要: Through bioinformatics and RT-PCR, in rice genome there were 8 VDAC genes, which located on the chromosome 1,3, 5, 9 respectively.In this project, through the anaysis for space-time peculiarity in transcription of the 8 candidate of the VDAC gene family in rice genome, the relatonship between different VDAC genes and the growth and development of rice will be studied; through the analysis of the full cDNA sequence and the forecast of the protein, the special domain of the different VDAC protein will be compared; through the immunity hybridization between the special antibodis from different VDAC domain and organelle and its proteins in different tissues of rice, the different VDAC proteins will be located in the subcell. Through the analysis of the mitochondria membrane permeability and its permeability transition in the tissues which the VDAC genes was expressed, the relationship between different VDAC genes and the functions of the mitochondria will be studied; knocking down 1-2 special VDAC genes through RNAi, the effects of different VDAC genes on the growth and development of rice will be probed into. This project will clarify the indispensable and the biofunction of the VDAC gene family, guide the application in the practice.
