中文摘要: 室性心律失常是心梗后常见并发症,致死率高。心律失常属于中医心悸、怔忡范畴,阳气不通是本病发生的关键环节,温通阳气是治疗本病的有效方法。葱白性温味辛,具有辛温通阳的功效,葱白提取物是葱白二氧化碳超临界萃取得到的有效部位。前期研究证实葱白提取物具有良好的心肌保护作用。预实验发现,葱白提取物鼻粘膜给药能够有效控制药物诱发的室性心律失常。本研究构建SD大鼠心梗模型,并诱发室速,评价葱白提取物的疗效,并评价其电生理机制。在此基础上,利用芯片筛选可逆性差异性表达的miRs,利用QRT-PCR验证,通过上调或下调相关miRs证实其功能和电生理相关性,预测靶基因,luciferase和上调下调实验证实目的蛋白的相关性,阐明葱白提取物-miRs-蛋白调控通路。本研究拟旨在探讨葱白提取物对心梗后室性心律失常的治疗作用,并从心电生理和miRs两个水平探讨其起效机制,为临床应用提供实验依据和理论基础。

英文摘要: Ventricular arrhythmia is a common complication after myocardial infarction, accompanied by a low survival. Arrhythmias is considered to be palpitation in Chinese medicine, and the obstruction of yang is thought to be the key etiology. Warming yang is thus an effective method in treating arrhythmia. Fistular onion stalk is characterized by warm and tasted acrid, and known to be effective in warming yang. Fistular onion stalk extract is the active part obtained from carbon dioxide supercritical fluid extraction, our previous study has confirmed that the fistular onion stalk extract had myocardial protective effect, and the preliminary experiment has shown that the extract is effective in controling the drug-induced ventricular arrhythmias by nasal administration. In this study, we will construct SD rat myocardial infarction model, induce ventricular tachycardia, observe the protective effects of fistular onion stalk extract, and evaluate the electrophysiological mechanisms. We will then screen the reversibly differentially expressed miRs using microarray and validate using QRT-PCR. We will next confirm their functions and electrophysiological relation through miRs up and down regulation assay. We will further predict the target genes by TargetScan or PicTar, and confirm its involvement by luciferase assay and up and down regulation experiments, and illustrate fistular onion stalk extract-miRs-proteins pathway. This study aims to explore the effect of fistular onion stalk extract on ventricular arrhythmias after myocardial infarction, investigate the electrophysiological and miRs based mechanisms, and provide experimental evidence and theoretical basis for its clinical application.
