中文摘要: 石墨烯的聚合物功能化和金属蛋白的生物传感研究是近年来的研究热点。本课题旨在将石墨烯的聚合物功能化、金属蛋白的生物催化特性和电化学传感技术相结合,在基质电极上可控修饰一体化的电化学生物传感膜,创建高灵敏度、高选择性、高电化学活性的生物传感新方法。首先采用具有特定功能的聚合物对石墨烯进行可控化学改性,获取具有高分散性、生物相容性和金属蛋白识别能力的功能化石墨烯。进而研究功能化石墨烯与金属蛋白的复合机理并进行传感膜的构筑与优化。最后利用金属蛋白的生物催化特性研究电化学传感膜的分子识别原理、响应特性及换能机制等生物传感的基本理论,建立痕量生物分子的简便、快速、准确的检测方法或疾病诊断体系。本项目的实施将揭示金属蛋白在新型石墨烯材料体系里的直接电化学和电催化传感机理,既有利于构筑更加广泛的石墨烯电化学传感器,又可为生物分子的高效检测开创一条新思路。

英文摘要: Recently, the study regarding polymer functionization of graphene and biosensor of metalloproteins have attracted much attention. This proposal is to investigate the combination of polymer functionization of graphene, biocatlysis of metalloproteins and electrochemical sensing with an aim to construct controlled modified electrochemical sensing films and develop novel biosensing methods with high sensitivity, excellent selectivity and electrochemical acivity. Firstly, polymers with special properties are utilized to functionalize graphene and obtain graphene with high dispersion, biocompatibility and recognition of metalloproteins. Then combination mechanism of functionalized graphene and metalloproteins will be performed with optimized structure of these biosensing membranes. Lastly, principle of molecular recognition, response characterization and transduce mechanism can be investigated to develop the novel electrochemical methods or disease diagnosis system for the simple, rapid and accurate detect of biological moleculars.
