中文摘要: 小RNA在植物的生长发育调控中发挥重要作用,Argonaute蛋白是小RNA介导的基因表达调控途径中的重要因子。本项目组前期基于突变体和RNAi的研究发现,ZmAGO18b可能参与玉米株高、雄穗长度和雄穗小花对数等性状的调控。本项目拟在前期研究的基础上,深入开展突变体和转基因后代的遗传分析,解析ZmAGO18b的生物学功能。同时,通过RIP-seq和CLIP-seq、RIP结合Northern 杂交或stem-loop PCR鉴定和证实ZmAGO18b特异性结合的小RNA;通过突变体与野生型的转录组比较分析,鉴定ZmAGO18b的调控途径、发掘小RNA的靶基因,RLM-RACE和QPCR验证靶基因。通过以上研究,以期建立ZmAGO18b-小RNA-靶基因-生物学性状间的联系,阐明ZmAGO18b的生物学功能和作用机理,为揭示AGO-sRNA沉默复合体在玉米生长发育中的作用机理积累有益资料。

英文摘要: Small RNAs play important roles in plant growth. Argonaute (AGO) protein is one of the essential components for small RNA mediated gene expression and regulation pathways. Our previous work founded that ZmAGO18b regulate plant height, tassel length and tassel florets pairs traits in maize based on the study of mutant and RNAi. In this project, we’ll assay the biological function of ZmAGO18b through genetic analysis of the ZmAGO18b::Mu and RNAi progenies. Moreover, an RIP-seq and/or CLIP-seq technique will be used to uncover ZmAGO18b specifically bound small RNAs and target genes that will be respectively validated by Northern blot, stem-loop PCR, RLM-RACE and Real-time PCR. Furthermore, gene expression difference will be revealed by RNA-seq. Together all of these results, we try to link ZmAGO18b, small RNAs, target genes and phenotype, and to elucidate the biological function and the mechanism of ZmAGO18b, then to accumulate useful information on mechanism of AGO-sRNA silencing complex function during plant development and growth.
