中文摘要: 线粒体铁蛋白(MtFt)是一种新发现的的定位于线粒体上的铁蛋白,其功能还不十分清楚,对其调控的分子机制也未有相关报道。我们前期的研究结果已经证实,MtFt通过维持细胞的铁稳态,对6-OHDA造成的神经细胞损伤具有明显的保护作用,有趣的是,在我们的前期研究中,发现了低氧对MtFt表达具有显著地上调作用,本项目从此出发,研究低氧对MtFt表达调控的分子机制和信号通路,在此基础上,利用MtFt基因敲除小鼠和转MtFt基因的神经细胞,研究MtFt对低氧引起的细胞损伤的保护作用及其机制。本研究不仅将首次阐明MtFt基因的调节机制,还将揭示MtFt对低氧诱导细胞凋亡保护作用及其新机理。

英文摘要: Mitochondrial ferritin (MtFt), a newly discovered ferritin, specifically expresses in high energy-consuming cells,including neurons of brain and spinal cord.However, little information is available about functions of MtFt. The regualtion mechanisms of MtFt is not known.Our provious study showed that MtFt plays an important role in preventing neuronal damage in an 6-OHDA-induced parkinsonian phenotype by maintaining iron homeostasis.Interestingly,we found that the hypoxia significant upreguated the MtFt expression in our previous study. The purpose of the present study is to evaluate the molecular mechanisms of MtFt regulation by hypoxia. We further explore its effects on hypoxia induced cell damage using the mouse of MtFt-/- , SH-SY5Y cells overexpressed MtFt. The present study will firstly identify the regulating mechanisms of MtFt by hypoxia, and provide a new advance in understanding the neuroprotective role of MtFt against the hypoxia damage.
