中文摘要: DNA结构及编码信息的变化是肿瘤恶性转化最根本的因素之一,通过检测这些肿瘤相关DNA在人体中结构和分布的变化,有望为肿瘤的早期诊断提供重要证据。本项目以活细胞中存在的G-四链体结构为靶向,通过设计构建同时具有结构识别单元及序列定位单元的双功能检测探针,实现对活体肿瘤细胞内目标G-四链体结构的特异性检测。本项目旨在建立一种活细胞目标G-四链体的检测方法,为开发基于肿瘤相关目标G-四链体的恶性肿瘤早期检测新方法奠定坚实的理论基础和有力的技术手段。

英文摘要: The research about the DNA structural alterations has been an important way to develop an efficient target for the tumor diagnosis or therapy in an early stage because the change of DNA structure and encoding generally is the origin cause of the tissue and cell canceration. This project plans to design a series of dual-functional G-quadruplexes probes which can be used to detect malignant tumor related G-quadruplex structures in living tumor cells. These dual-function probes that recognize target G-quadruplex in tumor cells with high specificity and sensitivity are comprised of structural recognition unit and sequence positioning unit. This project also wants to setup a novel method for detecting target G-quadruplex in living tumor cells and provide substantial basis and techniques to develop novel G-quadruplex based method for early detection of malignant tumors.
