中文摘要: 茉莉酸类化合物(Jasmonates,JA)是一种重要的植物内源激素,在植物的生长、繁殖和抗逆中起重要的调控作用,而植物如何感知环境信号并启动JA合成以及JA信号如何诱导下游抗性基因转录的分子机制还不十分清楚。组蛋白甲基化在调节基因表达等方面起重要作用,前期工作中,我们鉴定了一个受逆境诱导的水稻组蛋白去甲基化酶JMJ705,该基因能特异性去除抑制基因表达的组蛋白H3末端第27位赖氨酸三甲基化(H3K27me3)修饰;超表达该基因提高了水稻对白叶枯病菌的抗性,初步分析发现该基因正调控了部分茉莉酸合成以及逆境响应基因的表达水平。为了更深入的研究JMJ705在逆境和JA信号传递途径中的功能,本项目以JMJ705的超表达和突变体为材料,利用高通量测序分析等技术,分析 JMJ705在水稻中的全基因组结合位点,以及茉莉酸诱导下转基因材料中JA响应基因表达水平的动态差异和组蛋白H3K27me3修饰的变化

英文摘要: Jasmonates(JA)as an important plant endogenous hormones regulates many aspects of plant development, reproduction, and defence. However, the mechanism of how plants perceive various environmental signals to induce jasmonic acid synthesis and how JA induced resistance gene transcription remain unclear. Histone methylation is an important epigenetic modification in gene regulation, in preliminary work, we identified of a stress-responsive histone demethylase JMJ705 that specifically demethylates the repressive epigenetic marks H3K27me3 in rice. Over-expression of the gene enhances rice resistance to the bacterial blight disease pathogen Xanthomonas oryzae pathovar oryzae (Xoo). Further analysis revealed that JMJ705 positive control some JA biosynthesis and stress-responsive genes expression. In this project, we focus on identifying the function of JMJ705 on stress resistence and JA singnal transduction by the high-throughput approaches to study the genome-wide binding sites of JMJ705 and dynamic difference of gene expression and H3K27me3 level on JA-responsive genes in JMJ705 overexpression plants and mutants, to uncover the molecular mechanism of the epigenetic regulation in jasmonic acid signalling.
