中文摘要: 本项目针对临床上对脓毒症病原体早期诊断和预后判断的迫切需要,拟在我们原有的研究基础上发展新型的压电发射的毫米悬臂梁传感器阵列(PEMC)用于脓毒症多种相关标志物的快速、灵敏、高通量检测。PEMC传感器是一类由PZT和石英玻片构成的毫米尺寸的共振模式大悬臂梁。在实验室静态或动态的液相条件下,PEMC的高阶模式表现出的灵敏度高于目前报道过的任何一种生物传感器,可直接测量到fg级的质量变化。本项目拟采用不同毫米尺寸大小的PZT 和玻片以及不同设计方案制备新型PEMC传感器,并对影响PEMC灵敏度的因素进行系统的考察,利用抗体固定在传感器表面来获得脓毒症标志物检测的选择性和识别。并采用传感器集成技术发展灵敏度高、重复性好、操作简单、成本低的PEMC 传感器阵列,建立对脓毒症多种标志物的高通量、同时、快速检测新方法,以期形成该领域具有自主知识产权的关键技术,为脓毒症临床诊断作出新的贡献。

英文摘要: The overall goal of the proposed research is to develop a novel piezoelectronic-excited millimeter-sized cantilever (PEMC) array for the rapid, sensitive and high-throughput detection of multiple sepsis-related biomarkers that based on our previous research and the clinical urgent demand for early diagnosis of pathogens and prognosis of sepsis.The PEMC sensor is a resonant mode macrocantilever sensor that is composed of two layers: a PZT (lead zirconate titanate) layer and a nonpiezoelectric layer (quartz slide) of a few millimeters in length. PEMC exhibits high-order mode that is more sensitive than any reported biosensor under liquid immersion and flow conditions. In a laboratory setting it measures mass changes of femtogram. The first purpose of this project is to fabricate new PEMC sensors with different millimeter-sized PZT and slides as well as different design methods.Specially, the factors that affected PEMC sensitivity will be systematic investigated. The second goal is to convert the fabricated PEMC sensor into a format for field use. After antibody immobilized on the sensor surface, the sepsis-related marker will be selectively and identification.
