中文摘要: 肺癌是世界各地常见的恶性肿瘤之一。最近我们发现乙酰化在代谢中起着非常重要的作用。ACL直接影响脂肪酸从头合成的速率,是脂肪酸从头合成代谢的启动酶。研究发现柠檬酸裂酶(ACL)在肺癌等多种癌症细胞中高表达,提示该酶与肿瘤的发生发展有着密切的关系。对其研究主要集中在磷酸化修饰和转录调控上,而关于ACL的乙酰化修饰国际上未见报道。有意义的是我们前期工作发现肺癌细胞系中ACL受到乙酰化的调控, 随后的功能研究发现乙酰化可能调控其降解。基于ACL脂肪酸合成代谢的启动酶,而且ACL在大多数癌症细胞中高表达,同时对肿瘤细胞的发展又是至关重要的, 因此ACL是潜在的癌症治疗的靶标。研究乙酰化对ACL的分子调控机制及其功能,并用小鼠模型和临床肺癌样品验证其体内功能,从而阐明其与肺癌的关系。对其进行研究不仅可拓展对肿瘤发生及其Warburg 效应的理解, 而且可为肿瘤的治疗提供新的靶标。

英文摘要: Lung cancer is one of the most frequent malignant tumor in the world. Recently we found acetylation has essential role in cell metabolism. ATP-citrate lyase directly controls the synthesis rate of de novo lipid synthesis and catalyzes the first step of lipid synthesis. Studies have shown that ACL is overexpressed in various cancers including lung cancer, indicating the important role of ACL in cancer development. Previous studies have focused on phosphorylation and transcriptional regulation of ACL enzyme activity, while no report on acetylation regulation of ACL. Interestingly, our preliminary data found ACL is regulated by acetylation in lung cancer cell line A549, further study indicated ACL acetylation may control its degradation. Based on ACL is one of the important enzymes in de novo lipid synthesis, and ACL overexpression is founded in mutiple cancers and important for cancer development, ACL is a potential target for cancer therapy. Our study can provide not only the insight into cancer development and warburg effect but also the new target for cancer therapy.
