中文摘要: 偏头痛是一种难治性疾病,目前西医无法根治,中医药治疗偏头痛优势明显。正天丸是治疗偏头痛的首选中成药,但其止痛作用机制未明。中药复方作用机理的复杂性要求既从整体出发,通过系统的方法阐述其作用机理,又要从局部层次加以研究。本项目拟在原有工作的基础上,选择具有典型代表意义的偏头痛模型,一方面借助代谢组学采用UPLC-Q-TOF/MS技术对比正天丸治疗干预下动物整体内源性代谢物差异,探讨正天丸治疗偏头痛的宏观机制;另一方面,引入与神经病理性疼痛密切相关的ERK/c-fos蛋白通路,研究正天丸干预下细胞分子网络变化,阐述局部组织细胞学机制。最终利用生物信息学,将两者信息加以整合,从宏观和微观全面揭示中药复方正天丸的止痛机理。研究结果为中成药正天丸治疗偏头痛提供更充分的科学依据,对于中医药传统复方理论现代化有重要意义。

英文摘要: Migraine is one of the chronic and difficult diseases. In fact, Chinese medical therapy can obtain more remarkable curative effect than western medical therapy, and can meliorate the clinical symptoms of miagraine remarkably. Zhengtian Pill should be used as the first choice for clinical treatment of migraine as Chinese traditional medicine, but we do not know how it works. In order to investigate the pharmacological mechanism of Chinese traditional medicine, we should consider both global and local information. On the basis of the former achievement, we will use an ideal animal model to study migraine on the whole level by metabonomics technologies with UPLC-Q-TOF/MS and on the cell level by ERK/c-fos pathway, and then, integrate data by bioinformatics. The results of our project will provide scientific basis for the treatment of Zhengtian pill to migraine, and be important to modernization of Chinese medicine.
