中文摘要: 近期研究表明,血管支架对血管组织的成功修复需要同时解决抗凝血、促内皮细胞生长和平滑肌再生的问题。为了保证内皮细胞和平滑肌细胞都能快速的在管状支架上顺着各自的生理方向生长,促进血管组织再生,制备具有抗凝血、取向、大孔等特性的支架是急切需要的。本课题通过特制的纤维收集装置,以含聚2-甲基丙烯酰羟乙基磷酰胆碱的聚合物、聚乳酸-聚己内酯和聚环氧乙烷为材料,采用静电纺丝方法制备轴向平行纳米纤维内层和环向平行微米纤维外层的定向双层管状支架。将制备的支架溶于水中去除聚环氧乙烷,得到大孔支架。通过对支架的微观结构、力学性能、抗凝血性能、以及支架植入体内血流通畅性的研究,揭示定向双层支架对血管组织再生的方式,并做出一种能够最终用于临床的血管支架。

英文摘要: In recent, studies indicated that the success of the vascular tissue construction required scaffold with the performance of anti-coagulation, promoting endothelial cells growth and regeneration of muscle in the mean time. In order to ensure the smooth muscle cells and endothelial cells can be rapidly growth along with their physiological directions on the tubular scaffold, respectively, and ultimately promote vascular tissue regeneration, then, to prepare a functional graft with anti-coagulation, fiber-orientated, and macro pore is very necessary. For this project, an oriented, double-layered which with axial-parallel nano-fiber inner and circumferentially aligned micron fiber outer tubular scaffold will be fabricated by electrospinning X-poly (2-methacryloyloxyethyl phosphorylcholine) (X-PMPC), poly(L-lactic acid-co-ε-caprolactone) (P(LLA-CL)) and polyethylene oxide(PEO) with special fiber collection device. Upon fabrication,dissolving the PEO after immersing the scaffold into water to obtain macroporous structure. The scaffolds are evaluated with the microstructure, mechanical property, anti-coagulation property, and patency of blood flow in vivo to reveal the model of this structures potential to serve as a vascular prosthetic capable of in situ regeneration, and can be used for clinic ultimately.
