中文摘要: 抗生素的广泛使用,尤其是滥用,导致病原菌耐药性的问题已经成为全球关注的热点。发现结构新颖的具有协同抗耐药活性的活性化合物,使原来的抗生素"复活",是目前解决病原菌耐药性的重要策略之一。毛壳属真菌是一类重要的营腐生丝状真菌,其次级代谢产物结构类型丰富、活性种类广泛,目前尚无关于该属真菌中抗耐药活性成分的报道。我们在前期活性筛选实验中首次发现,毛壳属的4种菌代谢产物具有显著的协同抗耐药活性。因此本项目拟全面收集毛壳菌属的菌种,以耐药菌为指示菌,进行协同抗耐药活性菌种的筛选,获得10株以上具有显著协同抗耐药活性的菌株;利用HPLC-UV-HRESIMS联用技术及各种色谱分离手段,系统研究该属不同菌种的抗耐药次级产物,获得结构新颖的活性化合物;并对具有显著活性的化合物进行结构修饰及构效关系的研究;利用RT-PCR及二维凝胶蛋白电泳等分子生物学技术探索活性先导化合物的作用机制。

英文摘要: The widespread use,especially imprudent use of antibiotics in the treatment of different infections result in the global hot focus on the drug-resistant issue.To dicover new natural products with synergetic anti-drug-resistant activity is one of the most important tactcs to solve this problem.The genus Chaetomium is a class of important saprophytic mycelial fungi,whose secondary metabolites are characteristic of diverse structures with different bioactivities. Till now, there is no report on the secondary metabolites with synergetic anti-drug-resistant activities from the genus Chaetomium.In the process of screening for synergetic anti-drug-resistant metabolites from the Chaetomium species,the extracts of four species of Chaetomium genus exhibited significant bioactivities.In this project,we will collect the species of Chaetomium genus systematically, screen out more than 10 fungi with significant synergetic anti-drug-resistant bioactivities, investigate the bioactive secondary metabolites,study the structure-activity relationships of the bioactive compounds by structure modification,and elucidate the synergetic anti-drug resistant mechanisms of the lead compounds by using molecular biology methods such as RT-PCR and two-dimensional difference gel electrophoresis.
