中文摘要: 噬藻体被视为水体微生物群落的重要组成部分,其作为蓝藻特异性裂解因子,在蓝藻水华的形成与消亡过程中具有重要的生态学意义。本项目拟构建太湖浮游病毒基因组序列文库,应用生物信息学与分子生物学等技术,探明富营养化浅水湖泊中可能存在的噬藻体辅助代谢基因(与宿主代谢相关的功能基因)及其生物学功能;以鉴定的辅助代谢基因作为噬藻体的检测标记,针对天然环境中噬藻体的种群动态、时空分布及遗传多样性进行研究;在相关定量分析的基础上,运用多元回归方法分析噬藻体与蓝藻的种群结构变化关系,及其与环境因素之间的相关性。本研究将系统性解析富营养化浅水湖泊噬藻体的种群结构与多样性,探讨噬藻体对水华蓝藻种群结构演替及生理生化特性的生态影响,为开发噬藻体及其功能基因资源用于防控蓝藻水华和保护淡水资源提供理论依据。

英文摘要: Cyanophages have been considered as an important component of natural aquatic microbial communities, and are of great ecological significance due to controlling the formation and termination of cyanobacterial blooms as a lytic agent of cyanobacteria. In this project, based on a metagenomic sequence library constructed from Taihu Lake virioplankton, bioinformatics tools and molecular biotechnology are applied to assess the possible presence and biological functions of auxiliary metabolic genes (genes with putative functions in host metabolism) carried by cyanophage genomes from an eutrophic shallow lake; the identified auxiliary metabolic genes are used as biomarkers to reveal cyanophage population dynamics, space-time distribution and genetic diversities; and based on quantitative analysis, multiple regression method is applied to analyze the change relationships between cyanophage and cyanobacterial population strcutures, and the correlations between cyanophage populations and environmental factors.
