中文摘要: 流感如何获得跨动物间感染能力,使得从动物传播到人?人感染后是否会在人群中传播?这是当前预防医学领域中,人们最为关注和亟待解决的科学问题。 本项目拟开展以下三方面的研究:(1)创建细胞受体结构和功能的信息库;(2)流感病毒类型多样性和变异性的研究;(3)建立反向基因操作系统和多种原代细胞的感染模型两个技术平台。 本研究以细胞受体的特异性为突破口,创建并利用受体结构和功能信息库,建立原代细胞的感染模型和反向基因操作系统的技术平台,揭示受体结构功能的多样性与流感病毒变异的相互关系,探索并阐述流感病毒跨越宿主屏障能力的关键性分子基础。 预期研究结果将为人类流感预警预报、流感防治政策的制订提供直接的理论依据和技术指导,能进一步拓宽和提高人们对流感病毒的认知面和认知水平;为研制新型流感疫苗和抗流感病毒药物提供必要的科学依据;同时,在开展其它动物源性传染病跨种间传播机制的研究上具有重要的借鉴意义。

英文摘要: How does avian influenza virus get the ability of transmission from animals to human? And human infected avian influenza virus whether or not spread in crowd? These two questions are serious scientific topics in the currently preventive medicinal fields. In this study, we will take three works: (1) set up an information bank of various host cell receptor structures and functions; (2) research on the diversity and variability of influenza viruses; (3) establish two technology platforms of a reverse genetic system and some primary culture of several host cells. In this study, will first research the specificity of various host cell receptor, then set up and use the information bank of cell receptor structures and functions, establish the two technology platforms of reverse genetic system and the infect model of primary culture cell, clarify the interrelation between the diversity of the cell receptor and variability of influenza virus, and explore the key molecule elements of span host barriers' ability of influenza viruses. The expect research results will get direct theory and technical base for prevent and therapy influenza, great enhance cognize and knowledge of influenza virus, provide need basis for developing new type vaccines and drugs of anti-influenza virus, and they also get an important reference for investigating mechanisms of span host barriers of other pathogeneses from animals.
