中文摘要: 本项目将根据通过试验获取并经过预处理的基因活动状态(GAPs)时间序列建立一类参数不确定随机基因表达网络模型,并利用目前最前沿的随机分析工具和鲁棒控制技术对基因调控网络进行动态分析、性能综合和数值模拟。具体内容包括:对由DNA 微阵列技术获取的GAPs 数据进行预处理,建立判别GAPs 中存在误差和干扰的方法;根据GAPs 时间序列建立基因表达网络模型,着重考虑参数不确定性,并对随机噪音情况进行识别和处理;针对所建立的基因状态空间模型,进行鲁棒性能分析与综合,使得被控基因网络在有模型误差和随机扰动的情况下仍然能达到所期望的动力学行为;给出一个功能相容的计算软件包;参照大量真实的GAPs 试验数据,进行计算模拟和仿真并评价所建立的基因调控网络模型,进一步解释其生物背景和实际意义,以促进生物信息学科等相关领域理论和应用研究的发展。

英文摘要: This project will firstly construct a class of stochastic genetic network models with parameter uncertainties based on the gene activation profiles (GAPs) which have been acquired by experiments and pretreated before utilizing; then the dynamical analysis, properties synthesis and numerical simulations will be carried out on the genetic models by resorting to the most updated stochastic analysis tools and robust control techiniques. Specifically, this work includes: pretreating the GAPs data obtained by the DNA microarray techinques and establishing some critria for estimating whether there are some errors and disturbances existed in the GAPs; constructing the genetic expression network models based on the GAPs time series, here, parameter uncertainties will be emphasized and stochastic noises will be distinguished out and treated; robustly analizing and synthesizing on the constructed gene models so as to ensure the expected dynamical behaviors appear under the model errors and the stochastic disturbances; presenting a functional compatible computation software; based on the lots of real world GAPs data, simulating numerically and evaluating the constructed gene models, furthermore, explaining their biological background and the practical meaning to facilitate the theoretical and applied progress in biosciences.
