中文摘要: 本项目组前期研究明确了玉米大斑病菌Pathogenicity MAPK Cascade调控病菌分生孢子发育、附着胞形成、毒素的产生及致病力,并克隆了该条途径下游的转录因子STE12基因;生物信息学分析表明Ste12具Ste12-like特有的结构域,通过酵母双杂交技术明确了Ste12确与该途径的MAPK(Stk2)互作。本研究拟利用Real time-PCR、Western blot分析该基因在病菌致病过程中的表达模式;创制基因功能缺失和部分抑制突变体,通过分析突变体表型明确该基因与病菌致病性的关系;通过野生型及基因敲除突变体的转录组比较分析及染色质免疫共沉淀技术筛选Ste12作用的靶基因,进而创制靶基因的缺失突变体分析靶基因功能,明确STE12在玉米大斑病菌侵染过程中的调控机制。研究结果为深入探讨植物与病原物互作的分子机制和真菌病害防治的新途径奠定基础。

英文摘要: We identified previously that conidia development, appressorium formation, toxin production and pathogenicity of Setosphaeria turcica were regulated by pathogenicity MAPK cascade, then we cloned the gene STE12 of transcription factor which was located downstream of MAPK in this pathway. The protein Ste12 had a Ste12-like specific domain by bioinformatic analysis, further, yeast two hybrid experiments showed that Ste12 interacted with Stk2, MAPK in this pathway. In this research, we will analyze STE12 expression pattern in pathogenicity through Real-time PCR, Western blot; the knock-out and RNAi mutants will be constructed, and these mutants was used to study the relationship between STE12 with pathogenicity of S. turcica; its target gene will be screened through transcriptome comparison analysis between wild type and knock-out mutant strains and ChIP (Chromatin Immunoprecipitation), and the function of target gene will be characterized by gene knock-out. The regulation of Ste12 in infection of S. turcica will lay a foundation to study the molecular mechanism of plant-microbe interaction and provide a new approach of fungal disease control.
