中文摘要: 本研究拟通过免疫蛋白组学方法及免疫机理的研究,高通量地筛选具有良好免疫原性的斯氏艾美耳球虫候选抗原。通过二维电泳技术和免疫印记方法,分析具有免疫原性的蛋白;通过质谱分析与生物信息学分析,对具有免疫原性的蛋白进行分类鉴别及功能预测;通过动物免疫试验,评价候选抗原的免疫保护作用;通过测定肝脏几种重要酶的活性,揭示候选抗原对兔肝脏功能的影响;通过测定机体抗体水平,淋巴细胞增殖反应能力,细胞因子水平,以及T细胞亚类及B细胞水平,阐明候选抗原免疫机理。本研究将为兔球虫病的防制开辟新途径,为提高我国实验兔质量提供技术支持。

英文摘要: In this study, the immunoproteomic approach and the analysis of immunologic mechanisms were applied to screen potential immunogenic antigens of Eimeria stiedai. Two-dimensional electrophoresis (2-DE) and subsequent western blot technique were employed to analyse proteins of sporulated oocysts. The immunogenic antigens were identified by mass spectrometry and protein function prediction was performed by biological information analysis. The efficacy of immunization of potential antigens was evaluated by animal experimentation. The impact on liver was studied by detection of enzyme activity in liver. The serum antibody levels, lymphocyte proliferation responses, cytokines mRNA level and percentages of T lymphocyte subsets and B cells were determined to illuminate the immunologic mechanisms of the potential antigens. This study explores a new pathway for control of rabbit coccidiosis and provides technical supports to improve the quality of the laboratory rabbit.
