中文摘要: 受体和细胞微阵列芯片在生物医学、环境、食品等领域具有非常广阔的应用前景,是目前国内外生物医学传感技术的研究热点。本项目基于我们前期的工作基础,开展基于苦味受体和细胞微阵列的新型仿生传感器的研究。针对当前仿生传感技术亟待解决的若干关键问题,本项目基于无细胞蛋白表达系统和现代分子克隆技术研究功能性苦味受体和细胞的制备;基于DNA杂交编码技术研究受体和细胞与传感芯片的耦合机理,构建高度有序的特异性受体和细胞微阵列;基于差分进化算法优化仿生传感芯片的高通量快速响应特性。仿生味觉传感器不仅适合用于味觉传导机理的研究,也能用于特异性苦味物质的高通量实时检测。本项目的科学意义在于为基于受体和细胞微阵列的仿生传感技术提供新的理论依据和应用基础。尚未见到与本项目提出的仿生味觉传感器类似的报道,本项目具有较大的创新性和自主知识产权,有望在仿生传感技术若干关键问题以及味觉传导机理的研究方面获得一定进展和突破。

英文摘要: The receptor and cell micro array-based chips have shown widely promising applications in many fields such as biomedicine, environment, and food. Receptor and cell micro array-based chips have become a hot research topic in the field of biomedical sensing technology. This project will focus on the development of novel bitter receptor and cell micro array-based biomimetic taste biosensors on the basis of our previous research work. In order to address some urgent problems of biomimetic sensing technology, this project will study the production of bitter receptors and cells based on cell-free protein expression systems and molecular cloning. To construct highly ordered specific bitter receptor- and cell-based micro arrays, the coupling of bitter receptors and cells with sensing chips will be investigated based on DNA hybridization encoding technology. The diffenential evolution algorithm will be employed to study the high-throughput and fast response profiles as well as sensing mechanisms of biomimetic taste biosensors. The biomimetic taste biosensors can not only be used in the research of taste transduction mechanisms, but also be applied in the high-throughput and real-time detection of specific bitter substances. The scientific significance of this project is to provide new theoretical foundation and application basis for the receptor and cell micro array-based biomimetic sensing technology. There is no reports on the biomimetic taste biosensors similar to that proposed in this project. Therefore, this project shows significant innovation and independent intellectual property rights. This project has great potential to achieve distinct progress and breakthrough in the key problems of biomimetic sensing technology as well as in the research of taste transduction mechanisms.
