中文摘要: 造血干细胞(HSCs)的发育生物学研究具有重要的基础和临床意义。HSCs的细胞学起源已经有了定论,但它的分子调控机制还需要进一步研究。UBR5是一个具有多种细胞学功能的泛素连接酶。我们的前期研究显示,UBR5能够调控斑马鱼胚胎主动脉-性腺-中肾(AGM)区Runx1标记的HSCs的发生,在体外则能够上调BMP/Smad信号通路的活性。由于BMP信号通路直接调控Runx1的转录,在HSCs发生过程中必不可少,因此我们推测UBR5可能通过BMP信号通路调控AGM区HSCs的生成。本项目拟深入研究UBR5在斑马鱼HSCs发生中的生理功能,阐明UBR5调节BMP信号通路的分子机制,并探讨UBR5通过BMP通路调控HSCs 发生的机理。预期将鉴定和阐明调节BMP信号、通过翻译后调节方式调控HSCs发生的新分子和新机制,为HSCs发生发育的分子调控网络带来新的认识和研究思路。

英文摘要: Studies of the development of hematopoietic stem cells (HSCs) are significant for clinical and fundamental research. Recent studies in zebrafish convincingly demonstrate that HSCs are derived from the ventral wall of the dorsal aorta via an endothelial-hematopoietic transition (EHT) in the AGM region inside the embryo. However, the molecular mechanism of the formation of HSCs in the AGM region during embryonic development remains largely unknown. UBR5 is a ubiquitin ligase enzyme and possesses many cellular functions. Our preliminary studies showed that knockdown of UBR5 in zebrafish embryos repressed the emergence of HSCs, and that UBR5 upregulated the activity of BMP pathway in HaCat cells. Since the BMP pathway directly regulates the transcription of Runx1 and is required for HSCs emergence, we suppose that UBR5 is involved in the controlling the formation of HSCs through BMP patyway. In this study, we will further explore the novel role of UBR5 on the formation of HSCs, and reveal molecular mechanisms in the control of BMP pathway by UBR5. These results will provide new insights into the regulation of HSCs formation during development.
