中文摘要: 丙型肝炎病毒(HCV)感染的慢性化转归机理仍不十分清楚,不仅取决于病毒株的性质,还与宿主的遗传易感性相关。Th 细胞在 HCV 感染自限清除和免疫应答机制中发挥重要作用。Th 细胞表达的趋化因子受体影响 T细胞的极化结果,并通过与相应的配体结合,影响Th细胞亚群向炎症灶的募集和迁移。因此,Th细胞趋化因子受体及其配体的多态性,可能通过此通路影响宿主的免疫应答,进而影响HCV感染的慢性化转归。本研究拟在已有工作的基础上,利用既往有偿献血HCV感染高危人群队列随访资料,结合病例对照研究和分子生物学检测分析技术,分析HCV感染不同结局者Th细胞表达的趋化因子受体及其配体基因多态性和表达水平,探讨Th细胞表达的趋化因子受体及其配体基因多态性对Th细胞免疫功能的影响,以及主要SNPs与HCV感染慢性化转归的关系。

英文摘要: The infection of hepatits C virus (HCV) is one of the urgent public health problems recently. The mechanism of development and chronicity is still unclear. From recent researches, the outcomes of infection are related to both the characteristics of virus and the factors of host. Especially, Th cellular immune response performs a vital role in the process of spontaneous cl earance. Chemokine receptors and their ligands expressed by Th cell influence differentiation of T cells. Combining to ligands, chemokine receptors recruit the su bgroup of Th cells and migrate to the areas of inflammation. Therefore, the process also influences the outcome of HCV by the immune response of host. Associated with the expression levels of chemokine receptors and their ligands, this study focuses on the relationship between genetic variants of chemokine receptors, their ligands and the outcomes of HCV and focuses on the impact of immune function of Th cellular.
