中文摘要: 化感作用是多基因控制的数量遗传性状,会被环境因子所调控。植物在逆境环境下化感作用更明显,是其在进化过程中产生的一种对环境的适应性机制。前期研究结果表明伴生杂草可以诱导小麦化感物质的合成与释放,但对这一诱导效应的分子机制尚未阐明。本项目拟采用土壤盆栽共培体系,研究不同密度野燕麦胁迫下小麦化感品种京411和非化感品种百农矮抗58化感作用的差异,在此基础上,运用RNA-seq技术,生物信息学流程分析,阐明小麦化感物质代谢途径相关的差异基因,同时通过实时荧光定量PCR技术对这些差异基因表达行为进行验证,并应用UPLC/MS-MS技术检测小麦主要化感物质DIMBOA和MBOA的含量,解析伴生杂草野燕麦胁迫下小麦化感品种响应的分子机理,为代谢调控小麦化感作用提供理论依据。

英文摘要: Allelopathy is a multiple-gene-control quantitative genetic trait, and it will be regulated by environmental factors. Plant allelopathy is strongly enhanced under environmental stress, and it is an adaptation mechanism of plant to environment over evolutionary time.The previous research results showed associated weeds in wheat fields can induce the allelochemicals production in wheats, but the molecular mechanism of inducible effect is still not completely clarified. Firstly,in order to elucidate the change in allelopathic potential of wheat in the response to wild oat(Avena fatua L.)stress,allelopathic wheat "Jing 411" and non-allelopathic wheat "Bainong AK 58" are chosen as the donor plants and wild oat(Avena fatua L.)as the receiver plant in a pot and soil wheat/weed mixed culture system.Secondly,based on the previous results,RNA-seq and bioinformatics process analysis will be used to detect the differential expression genes which involved in the allelochemicals metabolism between the two wheat varieties under wild oat stress. Finally,quantitative real-time polymerase chain reaction (qRT-PCR) will be used to validate the expression of relevant differential genes.And the allelochemicals DIMBOA and MBOA contents in wheats will be determined by UPLC/MS-MS.
