中文摘要: 中国的东南部是全球鼻咽癌高发区域之一,对其转移机制的研究存在有強烈的需求。本项目拟凭借可视化光子技术研究鼻咽癌转移机制与LMP1癌蛋白的关联性,探索早期预测癌转移的潜在可能并进行调控干预,以提高患者的生存率。通过联合高分辨光学成像技术和荧光相关光谱技术,以鼻咽癌细胞脂筏微区所调控的LMP1蛋白及其介导的下游信号转导通路(PI3K/AKT)为特异性靶标,结合高灵敏度分子荧光探针技术,获取癌细胞脂筏微区的纳米分辨光学成像和LMP1在脂筏微区动态聚集特性的光学表征,以期认识LMP1对PI3K/AKT的募集特征和影响因素,达到可视化且半定量地预测鼻咽癌转移的倾向。本研究的预期目标是在细胞分子水平上获取表征鼻咽癌转移的光学图像与特征数据,将可为预测鼻咽癌浸润和转移潜能提供直观、可量化的参考依据和技术手段,实现早期调控并干预鼻咽癌扩散,达到有效地提高鼻咽癌患者治愈率提供新思路和理论支持。

英文摘要: Southeastern China is one of the highest risk area for incidence of nasopharyngeal cancer (NPC) in the world, there is a strong need on the research of metastatic mechanism of NPC. This project will study the correlation between LMP1 protein and metastasis of NPC based on visual Photonics Technology, and search for early prediction of potential cancer metastasis in order to improve the survival rate of patients. By combination of high resolution optical imaging technique with fluorescence correlation spectroscopy, to study the specific target of the LMP1 protein and the downstream signaling pathway (PI3K/AKT) which were regulated and mediated by nasopharyngeal carcinoma cell lipid rafts. Based on the high sensitivity fluorescence probe technology,we try to obtain the high resolution optical imaging of lipid rafts and the dynamic aggregation properties of LMP1 in lipid raft,and explore the characteristics and factors influencing PI3K/AKT collected by LMP1 in lipid rafts. Optical characteristics of nasopharyngeal carcinoma at the molecular level will contribute to predict the metastatic trend of nasopharyngeal cancer quantitatively, which will also provide novel ideas and theoretical support for treatment of nasopharyngeal carcinoma.
