中文摘要: 膝关节骨关节病(Knee OA)是临床常见疾病,流行病学调查发现维生素D缺乏与膝关节OA的发生密切相关,但机制尚不明确。为了研究活性维生素D在膝关节OA发生中的作用及机制,我们将分别给予同窝野生型和1α(OH)ase-/-小鼠高钙高磷饮食、皮下注射活性维生素D或高钙高磷饮食同时给予抗氧化剂NAC喂养;并建立1α(OH)ase-/-p16-/-双基因敲除小鼠模型,利用组织病理学、细胞生物学和分子生物学的方法比较分析上述各组动物的膝关节的表型差异,观察氧化应激、DNA损伤、细胞衰老、衰老相关分泌因子等指标的变化;并体外构建维生素D受体缺乏的OA软骨细胞,利用H2O2诱导其衰老,在培养中给予活性维生素D或NAC处理,比较分析各组细胞的表型差异。通过这些研究,以阐明活性维生素D缺乏导致膝关节OA发生的作用及机制,为维生素D在膝关节OA防治中的应用提供实验和理论依据。

英文摘要: Knee osteoarthritis(Knee OA) is a common disease.Epidemic investigation had demonstrated that vitamin D deficiency was associated with Knee OA,however,mechanisms are unclear.To investigate the roles and mechanisms of active vitamin D in preventing knee OA,1α(OH)ase-/- and their wild-type littermates will be fed on a high calcium and phosphorus diet,or administrated with active vitamin D or fed a dietary antioxidant NAC;Double 1α(OH)ase and p16 mutant mouse model will be generated;Phenotypes of knee from those animals,including alterations of oxidative stress,DNA damage,cellular senescence and senescence-associated phenotype will be analyzed and compared each other using histopathological,cellular and molecular techniches.In addition,VDR will be knock down in chondrocytes and these cells will be cultured with H2O2 in vitro,in absence or presence of active vitamin D or NAC and their phenotypes will be compared each other.These studies will be helpful to elucidate the mechanisms of the role of active vitamin D deficiency in inducing knee OA,thus will provide experimental and theoretical basis for the application of vitamin D in preventing knee OA.
