中文摘要: 洋葱(Allium cepa L.)为百合科葱属二年生蔬菜,首次在意大利红的品种中发现了雄性不育株后,由于其强杂种优势,F1代杂交种很快普及于世界各地。研究表明其不育性是由单个核基因和细胞质基因互作的结果。本项目利用我们课题组培育的雄性不育系和保持系为材料,采用实时荧光定量PCR和组织原位杂交的方法,对orf725嵌合基因进行差异表达分析,研究该基因的表达时期和部位;采用反向遗传学的方法,对orf725嵌合基因进行过表达和功能互补研究,明确该基因与洋葱细胞质雄性不育的关系。为阐明orf725基因的作用机理,揭示洋葱细胞质雄性不育分子机制提供依据。

英文摘要: Onion is a biennial vegetable, belongs to the Liliaceae family, Allium genus. The cytoplasm male-sterility (CMS-(S)) was discovered firstly in the onion cultivar 'Italian Red'.Male-sterile plants possess sterile (S)cytoplasm and are homozygous recessive at a single nuclear male-fertility restoration locus (Ms). The resource of this CMS-(S) are used worldwide as a male-sterile line for F1 hybrid onion varieties. In this project, two near isogenic lines of male-sterile and its maintainer line as the experiment material will be used.The real-time PCR technique will be used to research the specific gene expression stage and positions of the chimeric gene orf725, which caused by insertion of the alien fragment into the normal cytoplasm N. In order to explicit the relationship between the orf725 and male-sterility of onion, in situ RNA hybridization technique,gene overexpression, the functional complemention analysis of gene will be designed in this experiments to identify the gene function and action mechanism of orf725. This project will elucidate the molecular mechanism of male-sterility of onion,explore a new way to innovate onion germplam.
