中文摘要: 开发具有快速检测,高效吸附和生物应用等多功能的新型荧光探针,将是一项非常有意义的工作。经查阅近期研究成果发现,荧光探针仅局限于均相溶液中的应用,不能实现水相中对重金属离子的吸附与分离,存在生物兼容性和透膜性差瓶颈问题。鉴于此,拟设计合成系列新颖的喹啉酮-咪唑并[4,5-f][1,10]-邻菲啰啉钌配合物,将配合物固载功能化的SBA-15-NH2分子筛,首次构筑水溶性好的SBA-多吡啶钌配合物荧光探针(SBA-QIPAu),用吸(供)电基团修饰喹啉酮6-8位,调控体系的电子云密度,金属离子诱导喹啉酮内酰胺开环形成稳定的六员环螯合物,扩大共轭体系,增加质子在共轭体系内的荧光共振能量传递,产生荧光增强效应。相对于传统的荧光探针,本项目把探针的高选择性和灵敏度与SBA的生物兼容性和透膜性整合,同时兼备吸附修复与再生能力,预测探针可能实现多种癌细胞内的荧光成像,为开发新型多功能荧光探针提供技术指导。

英文摘要: To develop the new and multifunctional fluorescent sensor with fast detection,efficient adsorption and biological application is a very meaningful work. By reviewing recent research results, we found that fluorescent sensor can only be used in homogeneous solution, can't adsorpt and separate heavy metal ions in aqueous phase, and possesses the bottleneck problem of poor biological compatibility and cellular permeability of fluorescent sensor. Thus, a series of 2-[n-(R)-2-quinolinone-3-yl]-1H-imidazo[4,5-f][1,10]phenanthroline ruthenium complexes will be synthesized, the fluorescent hybrid materials SBA-polypyridyl rutheniumcomplex may be constructed by means of immobilization of fluorescence probewithin the channels of the functional SBA-15-NH2, in which 6-, 7-, 8-quinolinone can be connected by electron-withdrawing (donating) groups changing electrondensity of the conjugated system; lactam group of quinolinone is induced tocause ring to be opened and to cause chelate to be formed by heavy metallic ion, extending conjugated system, adding fluorescence resonance energy transferof proton in conjugated system, causing fluorescence enhancement. Compared with traditional fluorescent sensors, we can integrate best stability and sensitivity of fluorescent probe and excellentbiological compatibility and cellular permeability of SBA-15, and possess adsorptive and reprocessing ability. Biological imaging in many tumor cells may be predicted, which will provide a technical assistance for developing an novel and multifunctional fluorescent sensor.
