中文摘要: 肺癌是高死亡率的恶性肿瘤之一,其中又以非小细胞肺癌为主,难以早期诊断和治疗。核酸适体以其独特的优越性被认为有望成为新一代的分子药物,在生物医学领域显示了广阔的应用前景。本项目拟以申请人前期工作中已经筛选出的靶向识别非小细胞肺癌细胞系并可抑制其增殖的核酸适体为研究对象,确定功能性核酸适体的具体靶标及其结构特征;研究候选适体对非小细胞肺癌的增殖抑制作用相关分子机理和对肿瘤细胞的信号通路的影响; 研究核酸适体作为靶向载体对非小细胞肺癌细胞系的识别和药物输运的功能,为深入研究非小细胞肺癌及其它恶性肿瘤的致病机理,发展恶性肿瘤的核酸适体药物新技术打下基础。

英文摘要: Lung cancer is one of the high mortality rates of malignant tumors, especially non-small cell lung cancer. It is difficult to early diagnosis and treatment. Aptamer with its unique superiority is expected to become the next generation of molecular medicine and shows potential applictions in the biomedicl field. The project intends to use candidate aptamer from our previous work that can target and identify non-small cell lung cancer cell lines and inhibit cell proliferation as the target, determine the functioal aptamer's targets and their structural characteristics; researches related signaling pathways molecular mechanisms of tumor cell whose proliferation was inhibited by aptamers.Also, aptamer as kind of target vector could be studied. This project intends to research drug delivery for in-depth study the pathogenesis of non-small cell lung cancer and other malignancies, lays the foundation for the development of malignant tumors of the aptamers drugs as new technologies.
