中文摘要: 最近研究显示线粒体功能障碍是IUGR胎儿器官和组织(肝脏、肾脏、肌肉、脂肪)代谢异常的重要原因,肠道线粒体功能损伤可能是IUGR仔猪胃肠道功能紊乱的主要原因,但关于肠道线粒体形态和功能的研究未见报道。线粒体极易遭受自由基的攻击,而我们前期的研究发现精氨酸有抗仔猪氧化应激的作用。因此,我们假设精氨酸可以通过抗氧化应激途径缓解IUGR仔猪小肠线粒体损伤。我们首先以正常出生体重的仔猪为对照,比较研究IUGR仔猪小肠线粒体形态功能变化;然后采用不同水平精氨酸的人工乳饲喂3日龄断奶的IUGR仔猪,研究精氨酸对IUGR仔猪小肠线粒体的修复效应;最后,我们以H2O2诱导猪小肠上皮细胞线粒体氧化损伤,通过精氨酸和细胞抗氧化信号通路抑制剂处理,揭示精氨酸修复线粒体作用的分子信号机制。该项目的研究为深入认识IUGR仔猪胃肠道功能紊乱作用机制和精氨酸的营养功能积累资料,为科学饲IUGR仔猪提供依据。

英文摘要: Intrauterine growth restriction (IUGR) piglets have poor growth performance which is mostly related to gastrointestinal disorder. Recently, mitochondrial dysfunction has been proposed to play an important role in inducing metabolic disease of IUGR offspring, as indicated by the abnormal mitochondria function in internal organs such as liver, kidney, muscle and adipose tissue. However, it is unknown if intestinal mitochondria dysfunction exists in IUGR neonates. In the first study, therefore, naturally occurring IUGR and normal birth weight (NBW) pigs were selected to determine the differences in mitochondria function and morphology. Considering mitochondria is vulnerable to oxidative stress and the preventive effect of arginine (Arg) in oxidative stress, the second study was conducted to determine the recovery effect of Arg on intestinal mitochondria function of IUGR piglets. Thirdly, H2O2 was used to induce oxidative injury of mitochondria in IPEC-J2 cell line and the signal pathway of Arg in recovering mitochondria function will be investigated. These studies will reveal the molecular mechanism linking mitochondria function to gastrointestinal disorder of IUGR piglets.
