中文摘要: 人类转录组中存在大量的长链非编码RNA(lncRNA),它们在细胞中发挥重要的生物学功能。目前,需要对这些lncRNA进行鉴定和深入的功能研究。利用新鉴定的血管内皮细胞(VEC)自噬抑制剂3BDO处理VEC,基因芯片及生物信息学初步分析发现多个非编码转录本发生变化。前期,我们研究了来自TGF-β2 3’-非翻译区(3’-UTR)的lncRNA对VEC自噬的影响。但是,在VEC自噬受到抑制时是否还有其它lncRNA的参与并不明确。我们对芯片结果进行验证,结果显示一个可能是非编码的转录本-FLJ22182发生显著变化。本项目拟鉴定FLJ22182是否为lncRNA并研究其在VEC自噬中的作用。本研究可为阐明lncRNA调控VEC自噬的信号网络提供新证据,同时可为血管相关疾病的治疗提供新线索。

英文摘要: Long non-coding RNAs (lncRNAs) constitute a large portion of human transcriptome, some of these lncRNAs participate in important cellular functions. At present, it is necessary to identify these lncRNAs and further study their functions. When using a newly identified autophagy inhibitor, 3BDO to treat vascular endothelial cells (VECs), through cDNA microarray assay and bioinformatics analysis, we found that some possible non-coding transcripts were changed. In our previous study, we investigated the impact of an lncRNA derived from the 3’-untranslated region (3’-UTR) of TGF-β2 on VECs autophagy. However, whether other lncRNAs involved in this process was unknown. We verified the microarray assay results and found a probably non-coding transcipt-FLJ22182 was significantly down regulated. This project plans to identify a long non-coding RNA FLJ22182 and study its mechanism in autophagy regulation in VECs. This study can provide new evidence for elucidating lncRNA signaling network in the regulation of VECs autophagy and new clues for treatment of vascular related diseases.
