中文摘要: 本项目以长期堆积成的富含重金属的城市固体废弃物中土著微生物群落为研究对象,以用于协同浸出印刷线路板重金属Cu、Pb、Zn、Cr等的单株菌淋滤浸出模式和混合菌淋滤浸出简单模式群落为对照组,运用DGGE、Realtime-PCR和基因芯片等生物群落基因分子分析手段,结合原子吸收、SEM、TEM、ICP、XRD等化学分析手段,对比研究单一菌株、简单模式群落和复杂群落浸出系统中微生物种群在正常生理和环境胁迫下细胞、分子水平上相互影响和群落浸出功能分子相互作用,确定种群动态变化和功能活动在细胞、分子水平上与城市固体废弃物重金属淋滤浸出过程中各环境因素之间的关联,揭示微生物群落在协同作用淋滤浸出过程的演替规律。该项目对指导城市固体废弃物微生物群落结构与功能的优化调控,构建高效种群配伍与高效浸出工程菌,解决城市固体废弃物微生物淋滤浸出难题具重大的意义。

英文摘要: The indigenous microbial communities in city solid waste, which are rich in heavy metals accumulated longtime will be studied in this project. The control group microbes, which are single strain and simple community will be used for bioleaching of the heavy metal in printed circuit board, such as Cu, Pb, Zn, and Cr. The gene molecular method such as DGGE, Realtime-PCR, and gene chip, as well as the chemical analysis such as AAS, SEM, TEM, ICP and XRD will be applied to study microbes, which are single strain, single model communities and complex communities in normal physiological or environmental stress, and to understand the interaction and influence of function gene on cell and molecule levels in order to try to determine the correlation between dynamic state of population, functional activity and environmental factors in leaching of heavy metal in city solid waste, and to learn the succession law of group microbes in process of synergistic leaching. This project could be used to optimize microbial community structure and function and to obtain the leaching engineering bacteria with high performance compatibility, solving the difficult problems about city solid waste microbes' low leaching rate.
