中文摘要: 微生物可产生种类繁多的次级代谢产物,其中最重要的是抗生素。在目前所知的天然抗生素中约60%是由链霉菌产生的。谷氏菌素是由禾粟链霉菌产生的一种核苷肽类抗生素,该抗生素不仅对革兰氏阳性细菌(如金黄色葡萄球菌,藤黄八叠球菌)和阴性细菌(如大肠杆菌,普通变形杆菌)具有抑制作用,而且还具有抗植物病毒(如烟草花叶病毒,黄瓜花叶病毒)的作用。本申请项目以谷氏菌素产生菌-禾粟链霉菌为研究材料,在已克隆到谷氏菌素生物合成完整基因簇(GenBank JQ 307220)和已异源表达的基础上,对其中24个结构基因编码的酶在合成途径中的作用进行深入的研究,阐明合成途径,揭示基因的相互作用,基因转录与抗生素生物合成的关系。开展上述研究旨在阐明合成机制,为谷氏菌素的途径优化改造、产量提高以及为核苷肽类抗生素组合生物合成的理性设计等奠定重要的基础。

英文摘要: Microorganisms can produce a variety of secondary metabolites, of which the antibiotics are most important. So far as we know, about 60% of the natural antibiotics are produced by Streptomyces. Gougerotin produced by Streptomyces graminearus is a peptidyl nucleoside antibiotic. It shows potent inhibitory activities against both Gram-positive bacteria (such as Staphylococcus aureus, Sarcina lutea) and Gram-negative bacteria (such as Escherichia coli, Proteus vulgaris ),as well as plant viruses (such as TMV, CMV). Gougerotin producing strain-Strpetomyces graminearus will be used as research material. Based on accomplished cloning and heterologous expression of gougerotin biosynthetic gene cluster (GenBank JQ 307220), this project is proposed to study the functions of proteins or enzymes encoded by 24 structural genes in the cluster, and to elucidate the biosynthetic pathway of gougerotin, to reveal gene interactions and the relationships between gene transcription and antibiotic production. All the studies mentioned above are aimed to elucidate biosynthetic mechanisms and provide important data for the optimization of gougerotin biosynthetic pathway, the improvement of the antibiotic yield and the rational design of combinatorial biosynthesis among peptidyl nucleoside antibiotics.
