中文摘要: 多糖因参与病毒感染、免疫应答等生命活动而成为现代生物学和化学共同关注的焦点,但其高活性单体及生物标志物方面的研究尚未见报道,这源于多糖代谢物分析检测技术的局限以及当前多糖代谢研究策略的缺乏。本项目聚焦于红薯多糖的高活性单体,融合代谢组学的研究思想,形成糖代谢组学的新研究方向;设计同位素标记的质谱、荧光双增敏多功能分子探针,满足组学研究对多平台检测的技术需要;根据多糖活性的构效关系,将其代谢物划分为若干代谢组,结合"虚拟靶向代谢组学"以及"同位素标记"技术对各代谢组进行梯度式研究,形成基于同位素标记的糖代谢组学多平台梯度研究的新策略。再借助多变量方法优化标记条件与数据处理,建立标志物的预测模型。新策略的构建可提高样品检测的灵敏度、数据采集的准确性、标志物发现的概率与可信度,并可延伸至标志物的评价与预见。项目的实施将促进糖化学与代谢组学向更深处发展,对我国高附加值多糖产品的开发有着重要意义。

英文摘要: Polysaccharides have currently drawn the common attention of biologist and chemist for their participation in virus infection, immune response. However, the components of active single polysaccharides are still not clear and no reports on their biomarker discovery. These deficiency may be caused by the lack of analytical technique for polysaccharide and the limitation of research methodology for polysaccharide metabolites. In combination with the metabolomics, this project fouses on the sweet polysaccharide and its metabolites, forming the new branch "Saccharide Metablomics". Series of multifunctional(enhancing of Mass spectrum and Fluorescence signal) isotopical molecular probes (MIMP) will be designed to meet the requirment of metablomics. According to the structure-activity relationship, the metabolites will be devided into several groups and labelled with the MIMP, allowing for the new strategy of saccharide metabolomics. The labelling condition and data proceding will be finished with multivariate method, and also the prediction model for biomarker will be establsihed. The development of the new strategy will improve the analytical sensitivity, the accuracy of data collection, and the probobaility and reliability for biomarker discovery.
