中文摘要: 早期诊断癌症最有效的方法是通过肿瘤标志物的分析来判断,传统检测肿瘤标志物的方法存在灵敏度不高、易造成假阴性和假阳性、存在放射性污染、半衰期短、不易测量等缺点。本项目拟利用功能化介孔二氧化硅纳米材料包载量大、比表面积高、表面易于修饰、孔径可连续调控和水热稳定性良好等诸多优点,针对肿瘤标志物的电化学生物传感展开分析检测基础研究。以三种重要肿瘤标志物甲胎蛋白、蛋白激酶、甲基化酶做为检测目标,结合功能化介孔二氧化硅纳米材料的优良特性发展一系列准确、灵敏、快速的诊断方法,并在血清、血浆以及其他体液等复杂生物环境中考察并优化电化学生物传感器的性能。本项目的实施可以为癌症的早期诊断和早期治疗提供实用、有效的检测方法,具有一定的理论意义和重要的应用价值。

英文摘要: Early diagnosis and early treatment are critical to reduce the mortality. The most efficient approach is that cancer confirmation can be obtained by the analysis of tumor markers. However, the traditional methods of detection of tumor markers have several shortcomings such as low sensitivity, existing false negative and false positive results, radioactive contaminatiion, short half-life and uneasy detection. The project intends to take advantages of new functional mesoporous silica nanomaterials to analyze tumor markers in electrochemical biosensing. In order to achieve this goal, several important tumor markers, such as alpha-fetoprotein, protein kinase, methylase activity are used to the detection of cancer, combined with the excellent characteristics of functional mesoporous silica nanomaterials, a series of accurate, sensitive, rapid diagnostic methods are developed. After optimized the performance of biosensors, They can used in serum, plasma, other body fluids and other complex biological environment. The implementation of this project can provide practical, effective detection methods for the early diagnosis and early treatment of cancer, it also has certain scientific significance and the important application value.
