中文摘要: 石墨烯的制备是其工业应用的关键。目前的主要制备技术具有成本高、环境污染严重的缺陷。本项目将以氧化亚铁硫杆菌(Acidithiobacillus ferrooxidans,A.f)为出发菌株,经驯化、选育后获得高效氧化石墨菌种。采用该菌种对石墨进行多次生物氧化,绿色制备低缺陷生物氧化石墨(烯),拓展氧化石墨的制备方法。通过单因素、正交实验优化A.f菌9K培养基及路径,提高生物氧化石墨效率、减少黄钾铁矾矿物的生成量;以柱层析分离、纯化生物氧化过程中产生的酶,研究各种酶的性质及氧化作用,揭示A.f菌生物氧化石墨过程中的电子传递链及机理。优化石墨氧化条件,制备出高质量的氧化石墨(烯),为石墨烯的生物制备提供有效的技术支撑,并为了解生物氧化石墨过程提供重要的知识积累。

英文摘要: The preparation of graphene is vital to its industrial application. However,there are some defects on the current main preparation technologies of graphene, such as high cost and serious environment pollution. This item devotes to resolve such problems. To obtain the efficient oxidation strains of graphite, the Acidithiobacillus ferrooxidans(A.f) should be domesticated. Low defect microbial oxidation of graphite(graphene) can be green prepared through repeatedly oxidation by the A.f strains, expanding the oxidation preparation methods of graphite oxide. Single factor experiments and orthogonal tests of 9K medium and bio-oxidation process of A.f bacterial should be underway, improving the efficiency of microbial oxidation of graphite and reducing the formation of jarosite minerals. Separating and purifying the enzymes, which exist in the bio-oxidation process, by column chromatography. Nature and oxidation properties of these enzymes should be investigated and the electron transport chain and mechanism of graphite oxide must be revealed. Optimizing the oxidation conditions of graphite and producting high quality graphite(graphene) oxide, providing effective technical supportion and important knowledge accumulation as well as understanding to microbial oxidation graphene.
