中文摘要: 中医学肾主骨理论认为补肾法是治疗软骨退变性疾病的根本大法。基础研究也表明补肾药具有促进骨髓间充质干细胞(BMSCs)软骨分化的作用,但具体的作用机制仍不清楚。长链非编码RNA(lncRNA)是重要的表观遗传学调控手段,可多层次调控基因表达,影响干细胞的分化。根据前期研究提出科学假说:补肾药可能通过lncRNA促进BMSCs软骨分化。本研究拟阐明补肾药、lncRNA与BMSCs之间的内在联系, 有助于揭示干细胞软骨分化的表观遗传学调控机制,为应用补肾药联合干细胞促进软骨修复提供新的理论依据。 本研究通过lncRNA芯片、生物信息学、Real time RT-PCR、质粒转染以及RNA干扰等技术筛选左归丸和右归丸调控BMSCs成软骨分化相关的lncRNA,揭示lncRNA在左归丸、右归丸调控BMSCs成软骨分化中的表观遗传学机制。

英文摘要: According to the theory of kidney dominate bone in Traditional Chinese Medicine, nourishing kidney is the basic law of treatment of cartilage degenerative deseases.Basic research showed that kidney tonic herbs can promote bone marrow mesenchymal stem cells(BMSCs)to cartilage differentiation, but the specific mechanism is still not clear. Long non-coding RNA(lncRNA) is a new important epigenetic regulation tools. It can regulate gene expression at multilevel, influence differentiation of stem cell.According to previous research,we hypothesis that kidney tonic herbs may promote chondrogenesis of BMSCs by lncRNA. Clarifying the relationship between kidney tonic herbs, lncRNA and BMSCs, will help to reveal the mechanism of genetic regulation of stem cell differentiation, and promote the new theoretical basis for the repairing of cartilage with the application of kidney tonic herbs combined with stem cell. In this research, We will use lncRNA chip, bioinformatics, real time RT-PCR, plasmid transfection and RNA interference technology, reveal the epigenetic mechanism of lncRNA in chondrogenic differentiation of BMSCs reduced by Zuogui pill and Yougui pill.
