中文摘要: 食管癌是常见的消化系统恶性肿瘤,我国又是世界上食管癌的高发区。其传统的手术、放疗、化疗等治疗模式均存在着诸多弊端,目前学术界正尝试着采用分子靶向疗法治疗食管癌。由于具有易修饰、生物相容性好和形状可控等优点,DNA纳米结构正逐渐成为药物靶向输送的重要候选载体。本项目拟开展适用于食管癌分子靶向治疗的DNA自组装载药体系的建模及其载药性能研究。研究内容包括:通过设计、仿真和实验,构建DNA自组装基元类型库;基于Tile组装模型和Wang Tiles理论,利用DNA自组装技术,自底向上地设计DNA自组装结构体;基于最大熵和非线性系统理论,结合载药实证,通过数据整合和挖掘,自顶向下地建立DNA自组装载药体系的系统模型,并开展其可控性、适应性及载药性能研究。本项目的研究将为药物靶向输送提供新方法,同时有望为高生物相容性智能材料、纳米器件、高灵敏度特异性生物传感器等领域提供新思路和理论支撑。

英文摘要: Esophageal cancer, with a high incidence in China, is a common malignant tumor of digestive system.Researchers are now trying to apply molecular targeted drug therapy for esophageal cancer, because each of the traditional therapies (surgery, radiotherapy and chemotherapy) has its own disadvantages. DNA nanostructures, which have advantages of easy modification, good biocompatibility and shape controllability, are now gradually becoming one of the important candidate carriers for drug delivery. The modeling and drug release performance of DNA self-assemblied drug delivery systems for the treatment of esophageal cancer will be studied in this project.The main research topics of this project are as follows: Constructing the class libraries of the fundamental DNA self-assemble units by model design, simulation and experiment procedures; Bottom-up design of DNA self-assembled structures by DNA self-assembling based on tile assembly model and Wang Tiles theory; Top-down constructing the model of DNA self-assemblied drug delivery systems based on the theories of the maximum entropy and nonlinear system by data integration and mining, as well as combining with the drug-loading demonstrations; Studying the controllability, adaptability and drug loading property of DNA self-assembled structures.This project could provide not only new methods of targeted drug delivery, but also new ideas and theoretical supports for the high biocompatible smart materials, nano-devices, and high-sensitive and specific biosensors.
