中文摘要: 在肿瘤免疫治疗领域,多肽疫苗研发滞后于细胞疫苗和核酸疫苗。如何研发更好的能辅助诱导毒性T淋巴细胞增殖的佐剂,提高抗原肽的免疫原性,建立方便快捷的多肽组合方式是发展抗肿瘤多肽疫苗亟待解决的问题。本研究拟针对抗肿瘤多肽疫苗存在的上述问题,设计构建一种新型的抗前列腺癌多肽疫苗。该多肽疫苗基于霍乱毒素AB5分子结构,在同一大分子内同时包含细胞因子佐剂和五个抗原表位,各亚基借助霍乱毒素分子的自组装结构稳定结合且不相互影响,在解决抗肿瘤多肽表位免疫原性弱的基础上,能方便快捷的实现分子内多表位自组装,适应肿瘤个体化免疫治疗需要,且可利用霍乱毒素的黏膜佐剂特性实现无创给药。本研究在获得有价值抗肿瘤候选疫苗的同时,也对抗肿瘤多肽疫苗的设计提供有益的参考。此外,由于抗原肽和T细胞的相互识别是抗肿瘤毒性T淋巴细胞活化的第一步,对于该新型自组装多肽抗肿瘤疫苗的研究也将为细胞疫苗的研发提供重要参考。

英文摘要: In the field of cancer immunotherapy, peptide vaccine development lags behind the cell and nucleic acid vaccine. How to develop a better adjuvant to induce cytotoxic T lymphocyte proliferation, improve the immunogenicity of antigen peptide, establish a convenient way for polypeptide combination are the problems urgently to be solved for the development of antitumor polypeptides vaccine. This study aimed to design a new type of anti-prostate cancer polypeptides vaccine. The polypeptide vaccine structure is based on the AB5 molecular structure of the cholera toxin, containing a cytokine adjuvant and five antigen epitopes in the same macromolecule. The subunits within the macromolecule are formed a self-assembly structure, which is stable and let the epitopes do not influence each other. It can improve the weak immunogenicity of antitumor polypeptide epitopes, make it more convenient to change the composition of polypeptide vaccines. We aim to obtain valuable antitumor polypeptide vaccine candidates through this research, and provide a beneficial reference for the design of polypeptide vaccines against tumor. In addition, as the mutual recognition of the antigen peptides and the T cells is the first step of the activation of toxicity of antitumor T cell, this research will also provide useful information for the development of cell vaccine.
