中文摘要: 后发性白内障(PCO)是影响白内障手术效果的主要原因,其发生与晶状体上皮细胞(LEC)发生上皮间质转化(EMT)密切相关,目前对其发生机制的认识尚不充分。研究显示白内障术后晶状体周围产生电场,我们前期研究结果表明:电场作用下LEC发生成纤维细胞样变,细胞中E-cadherin表达下调,Vimentin表达上调,伴integrin表达升高和FAK的活化。以上结果强烈提示电场可能通过整合素相关信号通路诱导LEC发生EMT,进而参与PCO的形成。本项目拟在前期工作基础上,利用形态学、siRNA及相关分子生物学技术,从细胞和组织层面观察电场诱导LEC发生EMT和影响晶状体囊膜混浊的作用过程,以明确电场诱导的EMT在PCO形成中的重要地位;观察上述作用中整合素相关信号通路的调控能力并探明关键环节,进一步阐明电场诱导EMT及PCO的作用机制;结合以上结果观察药物联合干预的影响,为PCO防治开辟新途径。

英文摘要: Posterior capsule opacification (PCO) is the main reason that inhibit the vision rehabilitation after cataract surgery. The progress of PCO is related to the epithelial-mesenchymal transition (EMT) of lens epithelial cells (LEC), but the mechanism of it is still not well known.It is shown that an electric field (EF) is induced by cataract surgery. Our previous experiment data show that EF can induce the fibroblast-like shape change of LEC,and decrease the expression of E-cadherin and increase the expression of Vimentin,which including the activation of integrin and FAK. These results indicate that EF induces EMT of LEC by the integrin-related signaling pathway. In this reasch, with morphological and molecular techniques, we plan to observe the effect of EF-induced EMT of LEC and the effect of EF on the opacification of lens capsule, and to explore the role of integrin-related signaling pathway in this progress. By this reasch, we sought to clarify the role of EF-induced EMT of LEC in the formation of PCO, and to better our understanding of the pathogenesis of PCO. Combining with drugs which can manipulate the EF strengh, we strive to inhibit the EF-induced EMT, and search for a new way for the treatment of PCO in clinic.
