中文摘要: 慢性阻塞性肺疾病(COPD)是常见的慢性气道炎症性疾病,而皮质激素对COPD的治疗作用有限,本项目建立在我们补肾益气方治疗COPD临床有效并有所发现基础之上。补肾益气方能改善COPD患者肺功能、减少急性发作次数,降低COPD患者血清中的细胞因子水平,动物实验也发现能显著抑制COPD炎症反应。本项目首先构建香烟烟雾诱导的COPD炎症反应的动物模型和细胞模型,采用补肾益气方及其组分进行干预,并通过观察沉默PPARγ基因后补肾益气方及其组分对COPD炎症微环境的影响,评价PPARγ在补肾益气方及其组分抑制COPD进展的作用;其次从PPARγ影响COPD进展的已知途径入手,如检测HDAC2、GR及NF-κB通路等,探讨PPARγ在补肾益气方及其组分抑制香烟烟雾诱导的COPD炎症反应进展中的可能机制,为中医药防治COPD提供理论基础和新思路。

英文摘要: Chronic obstructive pulmonary disease (COPD) is a chronic inflammatory disease that is characterized by airway obstruction and progressive lung inflammation and is not significantly sensitive to corticosteroids. This study is established on the basis of effective treatment of BuShenYiQi granule (BSYQ), a Chinese herbs formula, on COPD and of previous research results. We found that BSYQ treatment ameliorated FEV1 (%) of COPD patients, reduced acute exacerbation frequency and down-regulated cytokines in the serum and the similar results were found in rodents experiments. This study first construct the cigarette smoke induced COPD models in vivo and in vitro, aiming to assess the role of PPARγ in the anti-inflammatory effects of BSYQ on COPD by silencing the PPARγ gene and treating with BSYQ. Furthermore, this study aim to explore the possible mechanisms how PPARγ affect the anti-inflammation of BSYQ on COPD from the known pathway that PPARγ affecting the progress of COPD, by testing the expression of HDAC2, GR and NF-κB in order to provide theoretical basis and new ideas for treating and preventing COPD of chinese herbs.
