中文摘要: 糖尿病是一种危害人类健康的重大疾病,临床血糖检验需要采血,且不能实现连续实时检测。中红外光谱对分子的选择性好,不同分子之间的光谱重叠和干扰不严重,信息丰富,所以人们考虑用中红外光谱进行无创血糖检测,但一直没有得到突破性的研究成果。其主要原因是由于人体皮肤是一复杂、时变的系统,其光谱易受到一些物理的、化学的因素的干扰。本项目研究了不同预处理方式对人体皮肤ATR-FTIR光谱的影响;结合模拟组织液样品,研究了人体皮肤ATR-FTIR光谱无创血糖测量方法中存在的化学计量学方法及光程变化、角质层的干扰等问题;确定了定量分析的模型,提出了校正光程变化的方法。由于人体ATR光谱主要反映的是角质层吸收信息,通过OGTT实验,没有得出人体皮肤的ATR-FTIR光谱与血糖之间存在相关性。我们认为,组织液很难渗透到角质层中,不宜通过ATR测量方式获取人体皮肤光谱来实现血糖含量的无创检测。

英文摘要: Diabetes mellitus,which menaced humankind's health, is a fateful disease. all present methods for glucose determination require patients to obtain blood invasively and unable to monitor glucose concentrations non-invasively. middle infrared (MIR)spectra gives more distinct absorption bands of each molecule and there is little disturbance between different absorption peaks.There are no satisfying results obtained from the glucose determination non-invasively using MIR spectra of human skin,the main obstacles are the changeable and complicated interference from cetain physical and chemical factors of human skin.in the project,the influence of skin preprocessing on MIR spectra of skin was studied firstly,and besides,we discussed the calibration method,optical path variartion and interference of stratum corneum(SC) coupled with the simulated interstitial fluid(ISF) samples.The spectrum of human skin is the only absorbance of SC just for the limited evanescent wave penetration,there was no correlation between ATR-FTIR spectra of skin and blood glucose concentrations in OGTT.It is concluded that ISF cannot easily penetrate into SC, ATR-FTIR spectra of skin is unsuitable for non-invasive blood glucose measurements.
