中文摘要: 皮肤组织因容易受到外界紫外线、化学物质的侵蚀而病变几率非常大。由于美容的因素,对皮肤组织的病理取样检查受到了很大限制。因此,实时无创功能学成像可大大提高皮肤病的临床诊断率。采用共焦光学技术可观察组织表面 200 微米深的细胞精细结构与生物化学成份,因而非常适合于皮肤疾病的病理诊断。本研究将采用两种模式对皮肤组织进行无创检测:利用近红外光共焦反射成像,提供细胞的折射率对比图像,对黑色素和角蛋白等组织结构进行成像;进一步通过共焦荧光层析成像,对线粒体自发荧光选择性成像,提取生物组织的新陈代谢和黑色素分布等生化信息。将这两种技术结合可实现功能成像,提高如皮肤肿瘤,银屑病,白癜风等皮肤疾病的早期诊断率。本项目将国际最新的技术前沿与申请人前期工作结合起来,实现组织的实时共焦三维功能成像。这一技术可取代传统病理取样检测,实现多种疾病的大面积无创实时检测,并利用功能成像实现疾病的早期诊断。

英文摘要: Because of the UV irradiation, chemical hazard, etc., the skin has a very high risk factor of disease. Due to cosmetic reasons, the histological analysis is highly restricted in skin diagnosis. Therefore, non-invasive, in vivo, real time functional imaging can dramatically improve the clinincal diagnostic accuracy. As confocal optical imaging can obtain cellular morphological and chemical information 200 um deep from the skin surface, it is highly suitable for dermatological virtual biopsy. Two modalities will be employed in this work for skin non-invasive diagnosis: with near-IR reflective confocal imaging, the refractive index contrast can be imaged, to reflect the structural information of the tissue such as melanin and keratin; with confocal autofluorescence imaging, the autofluorescence from mitochondria can be selectively detected, for the skin metabolic state as well as melanin distribution. Combining these two techniques, the functional confocal imaging can enhance the early diagnosis of a variety of diseases such as skin cancer, psoriasis, and vitligo, etc. This project combines the cutting edge technology with the experience of the applicant, to realize real time 3D functional imaging. The technique can take the place of conventional biopsy and histology, toward the non-invasive diagnosis of multiple skin diseases, and early diagnosis with functional imaging.
