中文摘要: P57是唯一被确认的非洲植物Hoodia gordonii中抑制食欲的有效成份,并被作为药物候选物研发了多年,但其作用机理一直未知。而其同系物gordonoside F是G蛋白偶联受体GPR119的特异性强效激动剂,在动物模型上表现出显著的治疗糖尿病效果。本项目将进一步合成扩展这类同系物,在此基础上制备相应的生物探针分子,以求确认P57的靶蛋白和gordonoside F与GPR119的作用机制。同时制备10克级活性化合物,用于动物药理实验。

英文摘要: P57 is the only confirmed appetite suppressant component in African plant Hoodia gordonii, and is being studied as a drug candidate for many years. However, its mechanism of action still remains a mystery. On the other hand, gordonoside F, one of the analogues, has been discovered as a specific potent agonist for the GPR119, and shows potent antidiabetic activity in animal models. In this project, more analogues and bioprobes will be prepared for determining the target protein of P57 as well as the mechanism of gordonoside F on GPR119. Meanwhile, the selected compounds will be synthesized in ten-gram scale for the pharmacology experiments on animals.
