中文摘要: 志贺氏菌(Shigella)俗称痢疾杆菌,是引起人细菌性痢疾(shigellosis)的主要病原体,其基因组、转录组、蛋白质组目前已得到了较为充分的研究,但是人们对它的蛋白质相互作用组(interactome)还知之甚少。主要挑战在于目前还缺少一种高通量、低成本的检测蛋白质相互作用的实验方法。随着生物信息学的不断发展,从蛋白质序列预测蛋白质相互作用组的计算方法日益成熟。本研究计划中,我们将以福氏志贺氏菌(Shigella flexneri)为模型,利用生物信息学的方法预测福氏志贺氏菌的蛋白质相互作用组,重点分析质粒编码的福氏志贺氏菌毒力因子在相互作用组中的分布特点,探讨质粒与染色体的相互作用,从而为福氏志贺氏菌致病机理的研究提供新的线索。

英文摘要: The genomes, transcriptomes and proteomes of Shigella, the etiologic agents of bacillary dysentery (shigellosis), have been investigated extensively but the protein-protein interactomes of Shigella spp. have not been addressed sufficiently. The challenge attributes to the lack of high-throughput and low-cost experimental methods for detecting protein-protein interactions. With the development of bioinformatics, computational algorithms for predicting protein-protein interactions based on protein sequences have been successfully developed. In this study program, we will use Shigella flexneri as a model and apply bioinformatic methods to predict its protein-protein interactome. We will analyze the structural properties of plasmid-coded virulence factors in the interactome to investigate the interactions between plasmid and chromosome, shedding light into the underlying mechanism of the pathogenesis of Shigella flexneri.
