中文摘要: 长链非编码RNA(lncRNA)与疾病的发生发展关系密切,但目前在心血管病方面的研究十分有限。我们的前期研究发现,细胞代谢的重要节点分子AMPK激酶的γ2亚基, 其编码基因PRKAG2的反义链存在lncRNA转录本,并调控该激酶活性,可能在高血压血管重塑过程中起重要作用。本项目以新发现的lncRNA酶活性调控机制对血管主要构成细胞的代谢及机能影响为切入点,从细胞、血管组织和机体三个层面阐明该反义lncRNA转录本对高血压血管重塑过程的影响及机制,并进一步揭示其在高血压病发生发展过程中的作用。项目成果对完善和推进目前lncRNA功能研究和AMPK激酶调控理论具有重要意义,也为防治高血压病这一棘手难题提供理论依据和解决方案。

英文摘要: LncRNAs are emerging as an important player in several human pathologies, but less progress has been made in the cardiovascular research. Our recent findings showed that an anti-sense transcript regulates the activity of AMPK, a critical mediator of metabolism, and may play an important role during vascular remodeling in hypertension. In this study, we will carry on our works from the impacts of the lncRNA on the function and metabolism of vascular endothelium and smooth muscle cells, further explore the mechanism by which the anti-sense transcript regulates the function of blood vessel, influences vascular remodeling process and the pathology of hypertension. We believe that the achievement of this project could remarkably enrich the current theories about functions of lncRNA and AMPK, and provide theoretical basis and solutions on overcoming the hypertension in clinic treatment.
